We propose to study specific manifestation of amide and disulfide groups
in solutions of peptides and proteins using Raman optical activity (ROA)
and electronic circular dichroism (ECD). Following the long tradition of
physico-chemical studies of biomolecules at the Institute of Physics of
MFF UK we are going to utilize the unique spectrometer for ROA
measurements. We intend to demonstrate the ability of ROA to provide
unique information on structural details of amide and disulfide groups
not readily accessible by other spectroscopic techniques.
The project is planned for two years with the expected ending by
December 31, 2013. In 2012 we will continue the interpretation of
already obtained spectra of the systems containing non-planar amide
groups (spirodilactams) and investigation of interactions of disulfide
bridges in short peptides (neurohypophyseal hormones or antimicrobial
peptides with one or two disulfides) The studies of more flexible
systems like short proteins and a correlation with theoretical methods
will follow in 2013.
