Raman optical activity of carbohydrates

Abstract: The work contains review about the state of art of Raman optical activity (ROA) of carbohydrates and glycoproteins. ROA provides an opportunity to study carbohydrate and protein moiety of glycoproteins in aqueous solutions independently. The study of monosacharides ROA spectra is essential for an understanding of ROA spectra of glycoproteins. In the experimental part we focused on ROA spectra of carbohydrates and we investigated spectra of D–fructose and both enantiomers of arabinose. Then we continued with D–glucose and L– and D–mannose, which has not yet been measured. In the cases of D–arabinose and D–mannose we tabulated positions of important spectral bands. This result allowed us to identify certain strong mannose bands in the spectrum of human orosomucoid. This work was a pilot study of measuring ROA spectra of carbohydrates and it demonstrated not only a suitable way of experimental procedures but also possible applications in the study of glycoproteins.
